Abstract
The planned research aims: 1) to unravel the nrITS, cpDNA and AFLP variability of the species within the 'Steveniella-Himantoglossum clade' with special emphasis on the species listed on international endangerment lists (Barlia metlesicsiana, Himantoglossum adriaticum, H. caprinum s.l.); 2) given the reported intra-individual nrITS sequence polymorphism in H. caprinum s.l., the research intends to test the method of sorting out the dominant cloned nrITS copies as orthologous ones. The research uses well-established phylogenetic methods for the conservational phylogenetic reconstruction, but it also extends our knowledge and methodology on orthology questions of nrITS. As possible outcome of the above, the applicant anticipates that this project will generate at least two peer-reviewed papers, together with a set of explicit recommendations to assist the conservation practice of key members of the Himantglossum group across Europe and Asia Minor. We reckon on the publication of the results of the first aim in a conservation journal and on a conservational conference, while the results of the second in a phylogenetic journal and on a evolutionary biology conference. Besides the scientific aim of the proposal, the applicant intends to join the leading-edge researches at 'Centre for Plant Diversity and Systematics' research group at University of Reading, and he would like to participate in the 'Intensive Course in Molecular Systematics' course for post-docs organised by the University.
